PF12172ChsH2, rubredoxin-like zinc ribbon domain (zf-ChsH2)ChsH2, rubredoxin-like zinc ribbon domainThis domain is found at the C-terminal end of the probable enoyl -CoA hydratase alpha subunit from Thermomonospora curvata (ChsH2) and similar archaeal and bacterial proteins. ChsH2 is likely to be involved in bile acid degradation. The C-terminal region of this protein is organised into two domains: an N-terminal zinc finger domain with a three-stranded antiparallel beta-sheet and an N-terminal alpha-helix (this entry) and a OB-fold domain (Pfam:PF01796) [5]. The domain is duplicated in some members like Swiss:O53566 from M.tuberculosis. The structure of Swiss:Q97WQ4 from Saccharolobus solfataricus reveals two long N-terminal helices followed by a rubredoxin -like zinc ribbon domain represented in this family and a C-terminal OB fold domain [1]. Zinc is chelated by the four conserved cysteines in the alignment.

ChsH2 is likely to be involved in bile acid degradation. The C-terminal region of this protein is organised into two domains: an N-terminal zinc finger domain (Pfam:PF12172) and an OB-fold domain (this entry) that consists of a five-stranded beta-barrel [6]. This domain is also found in 3-oxo-4,17- pregnadiene-20-carboxyl-CoA hydratase alpha subunit from Mycobacterium tuberculosis that is involved in cholesterol side chain degradation [3]. It is also found in Methanothermococcus thermolithotrophicus HMGCS, an enzyme complex comprising the thiolase and 3-hydroxy-3-methylglutaryl (HMG)-CoA synthase, that catalyses the second reaction in the mevalonate pathway-the exergonic condensation of acetoacetyl-CoA and acetyl-CoA to HMG-CoA [4].

PF00108Thiolase, N-terminal domain (Thiolase_N)Thiolase, N-terminal domainThiolase is reported to be structurally related to beta-ketoacyl synthase (Pfam:PF00109), and also chalcone synthase. Domain

PF22691Thiolase C-terminal domain-like (Thiolase_C_1)Thiolase C-terminal domain-likeThis domain is found C-terminal in a number of thiolases and their homologues. Thiolases are essential CoA-dependent enzymes in lipid metabolism. This domain folds into a four-stranded antiparallel beta-sheet with helices packed on one side. Domain
